In-vitro Anti-diabetic Evaluation of Sambirani Poo Kuligai by Alpha Amylase Enzyme Inhibition Assay

Abstract
Background: Diabetes mellitus (DM) is a metabolic disorder characterized by chronic hyperglycemia or increased blood glucose levels with disturbances in carbohydrate, fat and protein metabolism resulting from absolute or relative lack of insulin secretion. The Siddha system of medicine is a traditional medical system that uses a scientific and holistic approach to provide preventive, promotive, curative, rejuvenating and rehabilitative health care. According to Siddha literature, diabetes is known as Innippu Neer, Madhumegam and Neerizhivu. The various reasons for the cause of this disease are attributed to food, habits and lifestyle changes and also due to hereditary causes. The inhibition of alpha amylase enzyme involved in the digestion of carbohydrates can significantly reduce the postprandial increase of blood glucose and therefore can be an important strategy in the management of blood glucose level in type-2 diabetic and borderline patients.
Aim & Objective: This study aimed to evaluate the in vitro alpha-amylase inhibitory effect of Sambirani Poo Kuligai (SPK), a Siddha formulation, to assess its potential anti-diabetic properties.
Methods: Alpha-amylase inhibitory activity was assessed using a spectrophotometric assay.
Results: SPK exhibited alpha-amylase inhibitory potential, with a maximum inhibition of about 48.67% ± 4.097 and a corresponding IC50 value of 578.6 ± 44.75 μg/mL.
Conclusion: This in vitro study demonstrates that the Siddha formulation Sambirani Poo Kuligai (SPK) exhibits alpha-amylase inhibitory activity, suggesting its potential anti-diabetic properties.
